I am sorry about this but I am a real newbie boy on here, or maybe I am just a bit thick. I have chosen a theme called Mistylook by Sadish. It has 2 columns and 2 columns are set but I can not figure out how to use the second column and how to put something into that column. Please help.

I’d suggest you take a look at this tutorial:
http://learn.wordpress.com/The second column is meant to contain widgets which are little “gadgets” to help your readers see your recent posts, comments, links to blogs you like, categories and tags you assign to your posts, archives, and many other things. You don’t have to put anything into your sidebar (second column) but many of the widgets are quite useful.

That column is strictly for widgets. You install them here > Appearance > Widgets

Oh, one more thing, if you drag a widget into your sidebar (dashboard—>appearance—>widgets) and the others disappear, don’t worry. You can drag the ones you want back into the sidebar:
