  If i create a Hotel website and up grade to premium, can a guest book from my site and make payment through my site too? and can i register Email for my staff through my site???

  Unfortunately at WordPress.com you’ll not be able to receive payments for the rooms. Please take a look on Stay Theme, maybe it could help you.

  About Email registration, only with the Premium Plan you’ll not be able to register custom emails for your staff, to do this you have to buy a custom domain and map to your account, so you’ll be able to add emails for your staff.

  Partially, you’ll be able to create emails but you’ll not be able to receive payments, like I said before.
