how can i add tags to a tweet?

  I would like to add twitter tags to my tweet when I upload a blog post? Is this possible because it would help to publicise the post.

  Can I script the tween before publishing a post?

  The blog I need help with is: (visible only to logged in users)

  Sure. You can customize your message for your Publicize connections, as seen here:

  http://en.support.wordpress.com/publicize/#using-publicize

  In that customized message section, you can add your hashtags there.
