If I purchase a premium plan now, can I later upgrade to Business plan without changing anything in my site? If I need more than one sites of Premium level (as minimum), should I purchase two different Premium plans? Or Business plan includes multiple sites under the same plan?

You can upgrade to a Business Plan at anytime without changing anything on your website; however the Business Plan would open up additional features for the site in question. You would, however, have to pay the additional amount (i.e. the difference between the Premium Plan and the Business Plan) – you can read more about the WordPress.com plans here: https://wordpress.com/pricing/
Regarding your second question, please note that each plan is ONLY linked to one site. So if you need to have 2 websites on the Premium Plan then you will have to purchase 2 Premium Plans. I hope that answers your questions! Please let me know if you need any additional assistance :)
