Will I be able to have full control of the back end if I purchase a Premium or Business plan? Full control meaning editing of template files, uploading premium plugins and themes not hosted by WordPress, no limitations, etc.

https://move.wordpress.com/WordPress.COM and WordPress.ORG are completely separate and have different username accounts, logins, features, run different versions of some themes with the same names, and have separate support documentation and separate support forums.
