If I pay for the Personal plan (2.99), then decide to upgrade to Premum (8.25), will the money I spent on the annual Personal plan get deducted from the Premium? Or will I still spend the same amount that Premium is priced at?

Per the staff answer in the topic below, you will pay the difference between the two on the day you purchase the upgrade (prorated).

If, for example, you purchased a WordPress.com Personal Plan and 3 months later decide that you really needed the WordPress.com Premium plan, staff would apply a pro-rated discount to your Premium purchase. There are a couple ways staff could go about that, but the easiest would probably be to apply some credits to your account to reflect the discount. You would then purchase the Premium theme as you normally would, but at a discount due to the credits.

I’m afraid the information in the linked thread is not entirely accurate, as we don’t ever do pro-rated discounts or refunds on WordPress.com.
If you upgrade from a cheaper plan to a more expensive plan you pay the difference in cost between the two plans, and the expiration date is reset to a year from when you buy the more expensive plan.
