Hi there,
Generally speaking is not possible to set up a subdirectory structure like that when your sites are hosted with two different providers. Since my-site.com is with AWS and not WordPress.com, you can instead use a subdomain (like blog.my-site.com) to connect to the new site without changing anything about the current site. You can see the process in more detail here, but also we’re happy to walk you through it:
You can create a subdomain of any domain you own. This guide will show you how to connect a subdomain to your site on WordPress.com.About Subdomains
A subdomain has an extra piece of text before your main domain (called the root domain.) For example, if your root domain is yourgroovydomain.com, you can create subdomains such as blog.yourgroovydomain.com and shop.yourgroovydomain.com.

To be sure that I’ve understood your goals here, you are trying to find a way to add a blog onto your existing site since the platform it is built on does not provide that ability? Adding a blog to your site is a great way to improve your Search Engine Optimization (SEO) but you do not reap the full benefit by having your main site and your blog on two different website platforms.
With this in mind I’d also like to suggest that you consider moving the contents of my-site.com to WordPress.com as well. This will allow you to put the contents of that site on an SEO-optimized platform, and as a side benefit, it would make it possible to use my-site.com/blog as the URL for the blog page on your site.
In cases like that we recommend getting your site set up on the staging address we provide (both the main site and the new blog you wish to add) and then as a final step the domain can be connected seamlessly so there is no loss in traffic and everyone is redirected to the new site correctly.
If you are interested in hearing more about this we’re happy to chat about your needs and come up with an action plan that will help you to migrate smoothly.
Hope that helps. Please let us know if you have any more questions.
