Where did you get the
http://mansamusa.org.uk/home-2/URL? The 404 error you’re seeing at that URL means that particular page doesn’t exist.The page at your preview link appears to be the same as what shows on your site’s front page at http://mansamusa.org.uk/, so it looks like you have that page set as the static front page.

The Permalink settings you refer to control how permalinks on a site is structured, for example, whether post permalinks show the post date or not, or whether permalinks use the post/page ID or the post/page slug.
There is no possible way that changing permalink settings can resolve the 404 error – a 404 error means you’re using a link that refers to a page that doesn’t exist.
