Website Error HTTP 500 Internal Server Error

Need help quick. My employers business site is down completely and listing the subject line header:
WordPress is not even allowing me into the site to make any corrections, if that happens to be the case. We cannot have our site down for the whole weekend because that is where the bulk of our business comes from. Can someone please help me to figure out a resolution for this and if it is the case of billing, please let me know so that can be taken care of.
Also, because I could not gain access to the site, my password information did not work, therefore I had to make up a dummy account to write this since there is no listed number to call.

WordPress.COM and WordPress.ORG are completely separate and have different logins, features, run different versions of some themes with the same names, and have separate support documentation and separate support forums. Read the differences here http://en.support.wordpress.com/com-vs-org/
If you don’t have a username account at WordPress.ORG click http://wordpress.org/support/ and register one on the top right hand corner of the page that opens, so you can post to the support forums there.
