When using the theme customizer please be sure you are using one of the browsers and versions found here http://browsehappy.com and also be sure you have enabled third party cookies. http://en.support.wordpress.com/third-party-cookies/
Did you purchase a cusom design upgrade?
No changes you try out aside from those such as Custom Background, Custom Header and Theme Options which are stated on the theme description pages for each theme will be visible to visitors unless you purchase an annually renewable custom design upgrade?

Appearance > Header
Appearance > Background
Appearance > Theme Options (applicable only to some themes)
Settings > Reading (set a static front page)
Site Title and Tagline > Settings > General -
I’ve tried it on three of those browsers, and my cookies is enabled.
I did not purchase a custom design,
I had a blog theme which I could change and I changed which I would like to get back. Basically, I had pages with my posts under different categories. And now I which to have a ‘static page’ back. To change this before, I went on to customise, and changed it there. -
sorry I may have got that wrong,
‘static page’ I mean a normal blog layout where posts are in recent order -

re: front page error message
Oops! That page can’t be found.
It looks like nothing was found at this location. Maybe try one of the links below or a search?By default the front page of your blog displays all posts. The 404 (page not found) error message on the front page of the blog appeared because the Admin of the blog deleted the placeholder Hello World post. http://en.support.wordpress.com/not-found/ As soon as you publish a post, not a page it will disappear. Read the differences between Pages and Posts.

I’ve fixed my front page, and have my posts all on there now. But I still cannot customise my theme, or any theme the same way I used to. I am no longer able to press customise, preview my blog and use the design tool on the right hand side. They say they are loading but do not.

As timethief already pointed out, you don’t need the customizer. See here too:
