  If I purchase a premium wordpress membership, does that give me the opportunity to update themes with my own fonts and colours? And also, does the membership enable me to customise multiple blogs, or just one blog?

  Each upgrade at WP.com applies only exactly to what it says. That means each upgrade is PER BLOG. The Custom Design upgrade allows what you request, but only for the blog from whose dashboard it was purchased.
